requirements. We have vast experience in the analysis of biopharmaceutical
products to conform with
ICH guidelines and the FDA concept of a “well- characterised”
or “well-specified” molecule.
Applications range from simple molecular weight determinations
of peptides, to studies involving post-translational modifications
in recombinant proteins and carbohydrate structure in glycoproteins.
The M-Scan laboratories are equipped with a range of modern mass
spectrometers - 20 in total, linked to a variety of chromatographic
separation techniques.
Our ES-MS and MALDI-TOF services are capable of molecular weight
determinations on sub-picomole quantities of molecules. Structural
analysis can be performed using MS/MS or microbore LC-MS/MS.
The ‘MS-MAPPING’ technique involves
specific enzymic/chemical digestion of a protein, MS analysis
of the mixture of cleavage products and confirmation of assignments
by microchemical/enzymic techniques. We have applied this technique
routinely to the characterisation of recombinant protein/glycoprotein
products since 1982, even large antibody molecules. In addition,
this analysis can be extended to examine microheterogeneity at
both the N- and C-termini and to define the location of disulphide
bridges. These are areas which are difficult to analyse using
conventional techniques.
The most demanding high sensitivity sequencing studies, such
as Proteomics, are best performed using our recently installed
instruments of the novel Quadrupole Orthogonal Acceleration Time-Of-Flight
design, a Q-STAR and two Q-TOFs.
To complete our protein analytical capabilities we also perform
ancillary techniques such as SDS-PAGE (1D and 2D), gas phase sequencing,
HPLC and amino acid analysis (AAA) etc.
