Loke Diagnostics is specialized in production of specific bacterial antigens for use in ELISA-based assays and for production of specific antibodies. Antigen production is based on recombinant DNA technology and peptide synthesis. We are specialized in production of peptide antigens that have high binding affinity to ELISA plates.

Antibodies are produced by immunization with recombinant proteins or peptides. The antibodies are tested for reactivity with native microbial surface, for reactivity in paraffin embedded sectioned tissue samples, and for reactivity in ELISA and immunoblotting.

Development of subunit protective vaccine requires detailed information on immunogenic components. We use proteomics and immunoblotting of microbial antigens separated by 2D-PAGE reacted with patient serum samples to identify immunogenic components. The immune system is largely controlled by T cells where MHC molecules serve several important functions in the generation of a T cell immune response. We have implemented technologies to scan antigens for T-cell epitopes and use synthetic peptides to map specific CD4+ and CD8+ epitopes.

The peptide group synthesize peptides and improves current peptide synthesis procedures. The group has developed multiple peptide synthesizers which are used for synthetic work and for combinatorial chemistry. A new multiple antigenic construction, LPA (ligand presenting assembly) which is unique in providing simultaneous C to N and N to C oriented presentation of antigens of high homogeneity has been developed. Such peptides down to 8 amino acids has been shown to be immunogenic when used for immunization.
